1、盡可能是在蛋白表面 2、保證該段序列不形成α-helix 3、N,C端的肽段比中間的肽段更好 4、避免蛋白內部重復或接近重復段的序列 5、避免同源性太強的肽段 6、交聯可以交聯在N，C兩端，選擇依據就是交聯在對產生抗體不太重要的一端 7、序列中不能有太多的Pro,但有一兩個Pro有好處，可以使肽鏈結構相對穩(wěn)定一些，對產生特異性抗體有益。
